Mechanism of Action

Ibrutinib irreversibly binds to and inhibits BTK, a key enzyme in the B-cell receptor (BCR) signaling pathway.

• BTK is critical for the activation, survival, and proliferation of B-cells.

• In certain blood cancers, these B-cells become malignant and grow uncontrollably.

• By blocking BTK, ibrutinib disrupts cancer cell signaling, leading to reduced survival, migration, and proliferation of abnormal B-cells.

• This action causes cancerous B-cells to undergo apoptosis (programmed cell death) or become more vulnerable to immune clearance.

  Uses / Indications

  BD Brut (Ibrutinib 140 mg) is approved or used for the treatment of:

  1. Chronic Lymphocytic Leukemia (CLL) – with or without 17p deletion.

  2. Mantle Cell Lymphoma (MCL) – especially in patients who have relapsed after initial treatment.

  3. Waldenström’s Macroglobulinemia (WM)

  4. Marginal Zone Lymphoma (MZL) – in patients who have received prior therapy.

  5. Chronic Graft-Versus-Host Disease (cGVHD) – when other treatments have failed.

     Adverse Effects

     While ibrutinib is generally better tolerated than chemotherapy, it is not free of risks and requires regular monitoring.
